Evaluation of Double-Pipette Method for Recovery of Listeria spp . From Food Sample

Abstract
A recently-developed Double-Pipette (DP) method was evaluated for recovery of Listeria spp. from food samples. To construct the DP method, a 10 mL pipette was used as a container of universal enrichment broth for Listeria spp. Food or diluted food samples were directly added into a 10 mL pipette and mixed with enrichment broth. A three tenths mL of selective MOX (Modified Oxford) agar media for Listeria spp. was added into a 1 mL pipette and solidified. A three tenths mL of Brain Heart Infusion (BHI) broth was added onto the selective media through the top of the 1 mL pipette. A one mL pipette containing selective agar media and BHI broth was inserted into the 10 mL pipette to construct the combination of enrichment and selective procedures. After enrichment in the 10 mL pipette, the motile Listeria spp. moved up to the selective agar media and changed the medium color from white to black. After inoculation of motile L. monocytogenes into the food system (ground beef), the recovery of tested bacteria was evaluated using DP and conventional methods. The results showed that the recovery of L. monocytogenes was same for both DP and conventional methods. Also the efficiency of the detection for Listeria spp. from commercial food samples was same between these two methods. Therefore, the DP method is economical, rapid and simple for the recovery of motile Listeria spp. in food samples.
